Human Memory How reliable is Eyewitness Testimony This essays attempts to reach a conclusion regarding eyewitness testimonies and the extent of its reliability. This was done by identifying factors in three different processes stages. The acquisition stage is the period where the witness experiences the event. The retention stage is the period between the time the event ends and the time the witness are asked for a recount or questioned about the event. The retrieval stage is the period where the witness is providing information from their recall. During the acquisition stage, research regarding exposure time, estimation of factors concerning the event (for example, time), violence of the event, weapon focus and witness stress was analysed. During the retention stage, research regarding post-event information, introduction of conflicting information and introduction of misleading information was analysed. During the retrieval stage, research regarding method of questioning, leading questions and a case study of an actual event was analysed. Experiments and case studies regarding these factors were then evaluated to weigh the impact that it had on eyewitness testimony. In conclusion, it was found that experiments regarding eyewitness testimony hold little evidence to support its reliability but there are also some findings that dispute this. Thus, it was concluded that it is reliable only to a small extent though it can be highly accurate under certain conditions. Applications of this conclusion includes jurors being more aware of the factors regarding eyewitness testimony and taking into account when deciding on a verdict especially if the main evidence is based solely on eyewitness recounts. Eyewitness testimony is usually a verbal account given by an individual who has experienced an event, typically of a crime. Eyewitness testimony relies heavily on the capability of the individuals to accurately recount the event. In a trial, the jury is most often persuaded due to the statement(s) of the witnesses. Also, in cases where little material evidence can be collected, eyewitnesses are the focus for reaching a verdict. It has been argued that because testimonies are largely based on fallible memory which can be influenced by a variety of factors, it should not be depended upon. The contention of this essay is to identify and evaluate the extent of accuracy of this claim by identifying and evaluating factors that affect memory and by reviewing experiments and case studies to reach a conclusion on this matter. Memory and thus eye-witness reliability has become an issue of importance due to the high percentage of errors in identifying and prosecuting accused individuals. Thus, the reliability of eyewitness testimonies and impact on trials should be reviewed. Memory is defined as a kind of repository in which facts (information) may be retained over some period of time (Loftus, 1979). According to the Atkinson and Shiffrin model of memory storage (1971), memory is made up of three different types of information stores, each with different duration, capacity and function. The first is Sensory Memory. This type of store lasts for the spilt second when individuals collect information from their sensory systems and preserves information in its original sensory form. The sense organs are limited in their ability to store information about the world in an unprocessed way for more than a second. Thus, information is filtered through or selected by attention for further memory processing into the next type of memory store. This process of experiencing and filtering information is called acquisition. The next memory store is Short-term memory allows individuals to retain information long enough to be used and lasts approximately between 15 to 30 s econds. Miller (1956, cited in Weiten, 2005) proposed that Short-term Memory had a capacity of about 7 chunks of information. If certain actions are carried out, the information will be transferred to the last type of memory store, Long-term Memory. Long-term Memory provides retention of information which can last between minutes to a lifetime and has a limitless capacity. The process of information deposited in the short-term and long-term memory store is called retention. Information is constantly being transferred between these stores. When witnesses are asked to give recount of what they witnessed, information is taken from the long-term memory store and this process is called retrieval. Events during these three processes may affect the quality of the eyewitness testimony. During the acquisition process information may not have been perceived in the first place, information may be forgotten or interfered during the retention process or information may be inaccessible during questioning or during the retrieval process. Analysis of Factors that may affect Eye-witness Testimony during the Acquisition Process During the acquisition process, there are a number of factors that can affect an eyewitness report of an event such as exposure time or stress. This can be divided into event factors and witness factors. This section of the essay will evaluate the effect of these factors on the reliability of eyewitness testimony. Exposure time to the event or object of focus is an event factor. Laughery et al (1971, cited in Loftus, 1979) tested subjects on their recall based on the exposure time to a picture, showed one at a time of different positions of a human face. Two Caucasian male target faces were used, one with fair-colored hair and complexion with glasses and another with a darker-colored hair and complexion without glasses. The independent variable is the time the subjects viewed the pictures, which ranged from ten seconds to thirty-two seconds. The subjects were them asked, approximately eight minutes after exposure to identify the target within a series of 150 slides of human faces. The dependant variable is the accuracy of the subjects recall. Fifty-eight percentof the subjects who viewed the pictures for thirty-two seconds correctly identified the target but only forty-seven percent of the subjects who viewed the pictures for ten seconds correctly identified the target. This suggests that the more time a witness has to view the target, the more accurate their recall will be. This research is significant as it is very scientific and precisely tested a specific variable that affects memory and recall. Although this is so, it was also conducted in an artificial environment and thus, has low ecological validity. The research may also contain cultural and gender bias as it only tested for Caucasian males as the target. Thus, its application to targets of different cultures or gender is questionable. Estimating factors such as time, speed or distance is often asked of eyewitnesses. This involves perceiving the event and accurately inferring information from it. Marshalls (1966, cited in Loftus, 1979) experiment tested subjects estimation of time. Four hundred and ninety-one subjects watched a forty-two second film and a week after they had given their written and oral reports of the event, they were questioned as to the duration of the event. On average, subjects gave an estimate of about ninety seconds. The results show that witnesses can inaccurately estimate certain factors of an event. Although the results are significant, the study was conducted in a controlled environment which gives it little ecological validity. To further assess the accuracy of this study, an additional study that can be considered is Buckhout et al. (1975, cited in Loftus, 1979)s study on the effects if eyewitness testimony in a real situation by staging an attack where a student attacked a professor in front of 141 witnesses. The attack lasted for thirty-four seconds but when interviewed later on, the average estimate of the duration of the event was eighty-one seconds, almost twice the actual time. This study supports Marshalls study which shows that there is a tendency for witnesses to overestimate the duration in an event. This is significant in most cases, especially for cases of self-defense where the time between the attack and the retaliation is very significant in the categorization of the action. Another event factor is the violence of the event. A research done by Clifford and Scott (1978, cited in Loftus, 1979) investigated the ability of eyewitnesses to perceive violent and non-violent events. Forty-eight subjects with equal number of men and women watched either one of two tapes. In the non-violent version, the characters were involved in a verbal exchange and weak restraining movements. In the violent version, one of the characters physically assaults another character. In an effort to be even, the start and end of the tapes were manipulated to be identical. It was found that regardless of gender, the level of recall is significantly lower for those who viewed the more violent tape. It is inferred that this is due to the greater amount of stress that is produced in response to the violent event. This shows that eyewitness testimony of a violent event should be considered with the possibility of a higher rate of inaccuracy. Though the results are significant due to the hi gh reliability of the scientific method used, it also lacks ecological validity as it was conducted in an artificial environment where witnesses do not actually experience the event. An event factor that is linked to the witness factor, stress, is weapon focus. Easterbrook (1959) found that under high stress, individuals tend to concentrate more on a few features of their environment and less attention to other features. Weapon focus is where a crime victim is faced with an assailant who is brandishing a weapon. This rises the stress level of the crime victim and thus, they will only concentrate on a few features, mainly the weapon and will have trouble recollecting other factors such as the assailants features. Loftus et al (1987) tested this with an experiment where thirty-six students were showed a series of slides which showed one of two scenarios at a fast food restaurant. Half the subjects saw a customer pointing a gun at the cashier while the other half, the control group saw a customer handing the cashier a check. A part of the experiment tested the memory of the students based on a series of seven questions on the customer. It was found that the level of accuracy of the weapon group was fifty-six percent while the level of accuracy for the control group was sixty-seven percent. As the results show that the accuracy level of the weapon group is lower than the control group, this is highly significant in showing that eye-witnesses acquisition process might be seriously hampered by the presence of a weapon. The results of the experiment are highly valid as it was conducted in a controlled environment which allowed a direct cause and effect outcome. While this is so, the participants did not experience the event and thus their response may be different to an actual witness which means that the experiment lacks ecological validity. During an event, stress is a witness factor that should be taken into account. This refers to the level of stress or fear that a witness experiences which may influence their perception during the acquisition process of the event. A simulated case study done by Berkun (1962, cited in Loftus, 1979) placed army recruits in a stressful situation. They were isolated with the exception of a telephone link. Then, they were told that they were in danger to induce anxiety and were required to repair a broken radio by following a series of complicated instructions. It was found that the high level of anxiety impaired performance of the subjects. As this case study was conducted during a period of different ethical standards than today, there are ethical implications to be considered. Nevertheless, the results of this case study significantly support the Yerkes-Dodson law (1908, cited in Green) which states that emotional arousal facilitates learning and performance up to a point after which t here is a decrement. This can be applied to eyewitnesses who experience stress. Their senses may be stimulated but after a point, their acquisition process will be negatively affected. Although this is so, this case study has only looked at male soldiers, thus when applied to the general population, it lacks ecological validity. Analysis of Factors that may affect Eye-witness Testimony during the Retention Process Eye-witnesses are being tested on their retention of information from their Long-term Memory. Because retained information and thus, memory is being transferred between memory stores, it is possible that it can be influenced, enhanced or even distorted during or in between transfers. This section of the essay will focus on the numerous researches that have been carried out to investigate the accuracy of this or the extent of the influence that may occur. After an event occurs, sometimes witnesses discuss what they saw with each other and the information that is exchanged can result in an enhancement of memory in the form of modification or addition. An experiment was conducted by Loftus (1975) on one hundred and fifty participants. They were showed a film of a car crash where a car failed to stop at a stop sign and turned right to enter traffic, causing a five-car collision. After the film which lasted for less than a minute, participants were asked a series of ten questions. Half of the participants were asked about the stop sign in the first question while the other half were asked about the right turn. All other questions were the same. The independent variable here is the first question and the dependant variable is the last question which asked participants if they remembered seeing a stop sign. Loftus found that there was a higher percentage of participants who recalled seeing a stop sign if the first question related to a stop sign than the control group, fifty-three percent and thirty-five percent respectively. This shows that by mentioning an object, there is a higher chance of it being recalled. The results are highly significant as it was calculated using precise results with a specific variable tested. It can be argued that the experiment lacks ecological validity as it was conducted in an artificial environment but this allowed a specific variable to be tested which would have been impossible with a case study. As demonstrated by this experiment, eye-witnesses memory may be influenced by post-event information that they receive and thus, the more information that the witness is exposed to after the event, the higher the chance that their memory might be compromised. A modification to the previous variable is the question as to what happens when a witness learns new information which is different from what they experienced. Loftus (1975) conducted an experiment where forty participants were shown a three-minute video which involved a group of eight demonstrators noisily interrupting a lecture. The participants then had to answer a series of twenty questions. All the questions for the participants were identical except for one. Half of the participants were asked, Was the leader of the twelve demonstrators who entered the classroom a male? and the other half were asked, Was the leader of the four demonstrators who entered the classroom a male? All questions had to be answered with a yes or no. A week later, participants had to answer another set of questions. The critical question was How many demonstrators did you see enter the classroom? It was found that participants who previously answered the question with the word twelve reported an average of 8.9 demonstrators while the other half reported an average of 6.4 demonstrators. It can be argued that this experiment lacks ecological validity as it was conducted in an artificial environment and that participants only viewed the event and did not actually witness it. The results may not be fully applicable to the general population. Although this is so, the controlled environment showed a direct cause and effect of a specific variable. The results of this experiment can be applied to eye-witness testimony where witnesses may be mislead in their testimony as their memory might be compromised by the introduction of conflicting information. Similar to the experiment above is an experiment conducted by Loftus and Zanni (1975, cited in Hill, 1998) which was a modification of the original Loftus and Palmer research. The aim of the experiment was to investigate the effect of adding post event information on memory. Participants were shown a film of a car accident after which they had to answer a series of questions. Half of the participants were required to answer, Did you see the broken headlight which implied that there was a broken headlight while the other half were asked, Did you see a broken headlight which only required participants to recall if it was there. It was found that half of the participants, who were asked using the word the, incorrectly reported seeing a broken headlight. As most of the other variables were controlled in the experiment, and only a specific variable was changed, the results signify that memory can be modified by post event information. It can be argued that the participants did not experie nce the incident and thus the experiment lacks ecological validity when applied to actual witnesses but as a specific variable was examined, it can be concluded that the cause and effect was to a large extent connected. This experiment shows that eyewitnesses may be influenced by post event information which may negatively affect their account. McCloskey and Zaragaza (1985) also conducted experiments concerning the effect of misleading post event information on participants. After watching a series of slides depicting an event, participants received a narrative of the event. Participants in the mislead condition will receive a narration with receive the narrative with misleading information about a detail of the event, a hammer was referred to as a screwdriver instead while the control group werent provided details of the specific event. Participants then had to answer a series of questions about the event. The critical question concerning the tool had the original item as an option (hammer) and a new item (wrench). It was found that accuracy for the control group was seventy-five percent and seventy-two percent for the mislead condition group. The fact that there was little difference between the two groups points to the idea that misleading post event information does not distort memory of an event. This experiment suppor ts the validity of eyewitness testimony and due to the controlled setting; the results are highly valid though it lacks ecological validity. Analysis of Factors that may affect Eye-witness Testimony during the Retrieval Process Most evidence from eyewitnesses are a result of their accounts of the event. This involves the retrieval process of information from their long term memory store. This section of the essay aims to analyse and evaluate research conducted in relation to the techniques of retrieving information from eyewitnesses such as question wording or method of questioning. This is to evaluate the extent of influence of such techniques on the reliability of eyewitness accounts and in relation, memory. The method of questioning during the retrieval process plays an important role in the accuracy of eye-witness testimony as investigated by Lipton (1977, cited in Wells, 1978). Lipton conducted an experiment where participants watched a film of a murder in a courtroom setting. He found that unstructured testimony which allowed free recall resulted in ninety-one percent accuracy. This is a significant value as compared to other types of questioning such as open ended questions with eighty-three percent accuracy, leading questions with seventy-two percent accuracy and multiple choice questions with fifty-six percent accuracy. These results show that the type of questioning that the witness is subjected to affects the accuracy of their recount. When applied to trial testimonies, jurors should take into account the type of questioning to predict or get an idea of the level of accuracy of the testimony. Though conclusive to a certain extent, the experiment lacks ecological validity as the participants did not actually experience the event, merely perceive it but as it is a controlled environment, the reliability of the cause and effect factors is high. Loftus and Palmer (1974 cited in Hill, 1998) carried out an experiment to investigate the effect of leading questions on the accuracy of participants in recalling a car crash. Fourty-five participants were separated into seven groups and each group watched a video of traffic accidents. The videos lasted from five to thirty seconds. After watching the video, participants had to give an account of what they had just seen. The independent variable is the question About how fast were the cars going when they hit each other?. The word hit is replaced with the words smashed, contacted, bumped and collided for different groups. The participants answer as to the estimate of the cars speed is the dependant variable. Loftus and Palmer found that the mean estimate of speed for more aggressive words such as smashed is higher than less aggressive words such as contacted. The results are highly significant, p Contending the results of this experiment is Yuille and Cutshalls (1986) case study of a real life event. 13 participants were interviewed using Loftus and Palmers (1974 cited in Hill, 1998) technique in their recall four to five months after witnessing an attempted robbery in daylight where one individual was killed and another, seriously wounded. It was found that there was a very high level of similarity between the accounts given by the witnesses, the accounts did not alter in response to leading questions and that the witnesses were able to recall the event in detail. Additionally, accounts of those who were more distressed had a higher accuracy level. These results are different to Loftus and Palmer (1974 cited in Hill, 1998). The eye witnesses did not alter their accounts greatly in response to leading questions. As this is a case study, it holds high ecological validity unlike laboratory experiments. Although this may be true, Yuille and Cutshalls case study was of an event t hat was relatively traumatic event and was viewed in ideal conditions. Most incidents do not mirror this setting. Also, it was an investigation of only one case study. Hence, the application of these findings is debatable when applied to general eye witness testimonies. Conclusion This essay evaluated the extent of accuracy of the claim that eyewitness testimony should not be depended upon. This was done by focusing on the different factors that can affect human memory, and in relation, eyewitness testimony as it is fundamentally based on memory. Experiments and case studies related to factors in three different memory stages were identified and evaluated. Research concerning the acquisition process such as exposure time, estimation of factors concerning the event, for example, time, violence of the event, weapon focus and witness stress level during the event was analysed. Results point to a correlation of high levels of inaccuracy when more traumatic factors are included in the event. This is also reflected in research concerning the retention process and the retrieval process although there is some evidence that supports the reliability of eyewitness testimony. Overall analysis criticised the low ecological validity of controlled experiments but also supported the high level of validity that comes with it as it essentially provides a cause and effect relationship between specific variables tested. Case studies were found to be very specific with regards to certain factors, which leads to a debate about their general application. Since experiments are only replications of real-life events, it cant be fully taken as the actual process . This may have some influence on the results obtained as it can be argued that the participants are not really experiencing the event, thus essential factors like, atmosphere, or even interest in the event may be compromised. In this manner, results of case studies hold a higher level of reliability. As indicated by the numerous researches on different factors during the process of collecting, processing and retrieving information from memory, eye witness testimony is reliable only to a small extent. Under such fallibility, it can be questioned if eyewitness testimony should be relied on at all. Though eyewitness testimony has been proved accurate in a number of researches, the amount of investigations concluding on the fact that it is highly imperfect far outweighs it. Though a large number of researches do not support the reliability of eyewitness testimony, there is also evidence to support it like Yuille and Cutshalls (1986) case study. This suggests that although eyewitness testimony can be unreliable, under certain conditions, it is highly accurate. Implications for application of the analysis in this essay could include jurors being more wary of eyewitness testimony and the conditions relating to it when deciding upon a verdict. Due to the mediaââ¬â¢s brainwashing, poor self-image is one of the main causes of eating disorders in adolescent girls. The media sells products, not opinions nor facts. They will sell you anything, telling you that it will make you slimmer and that being skinny is the way to go. When we see models and celebrities on commercials and in movies, we often wish that that was us in that body, because the media has made everyone so obsessed with their own bodies. The media makes young female teenagers feel guilty if they are slightly overweight. Models are beautiful, skinny, they have the right size thighs, hips, and butt. They are models but they are not role models. They are everything teenagers want to be, because of the television they watch and the magazines that they read. The media are always telling them that being thin is considered beautiful. This sends a wrath between females and their impression of what beauty really is. Some females will go to any length to try to be that perfect body weight person. Such a person doesnââ¬â¢t even exist. For there is no perfect weight for which females should strive to achieve. Although there are many young females who will restrict what they are allowed to eat & when they are allowed to eat it. They will exercise compulsively and self effectuate vomiting. Eating disorders such as anorexia, bulimia, and compulsive overeating are three of the major eating disorders that infatuate young minds. Female teenagers see bingeing, dieting and vomiting, a way out of actually doing a workout and getting into shape. There is a widening gap between girlââ¬â¢s self-image and societyââ¬â¢s messages about what girls should be like. The media uses television, magazines and subliminal messages to attract people. They have playboy models advertising for cars, boats and body lotion. They use a womenââ¬â¢s body to advertise for their products. The media doe... ... they need to resolve, like the depression of not being happy with themselves. Of going through many procedures to make them look ââ¬Å"thinâ⬠or ââ¬Å"ordinaryâ⬠because they can no longer be happy with themselves because of the mediaââ¬â¢s interference with adolescent girls images. Anorexia and bulimia are one of the main causes along with the media as to why adolescent girls are always slightly underweight and devastating skinny. If the media didnââ¬â¢t interfere with adolescent perceptions, maybe one half of fourth grade girls wouldnââ¬â¢t be on a diet. Intense fear of becoming fat and distorted body images arenââ¬â¢t the kinds of thing we want our adolescents girls to become. To think about their appearance and weight 24/7 isnââ¬â¢t right. The media should stop publicizing young female bodies and telling them what beauty is perceived to them. They should think of all those adolescents girls that they send into depression or force to become bulimic, just because they wish to make money. The media brainwashes too many of adolescent girls, making them think about their body in relation to models who have unmistakably the ââ¬Å"perfect bodyâ⬠. Making them think that you have to be thin in order to be beautiful.

Evaluation of Genetic Diversity of Golden Apple Snail, Pomacea Canaliculata
ABSTRACT Genetics is a trend these days especially that, DNA barcoding has been developed. DNA barcoding is an important tool in categorizing the taxa of different species and it tells so much about the speciesââ¬â¢ traits, including genetic diversity. The Pomacea canaliculata was introduced in different parts of Asia and had been an invasive species and a pest in different ecosystems ever since the introduction. In understanding this species of snails, samples were collected, DNAââ¬â¢s were extracted, undergone PCR and electrophoresis, and was sequenced and analyzed.The analysis was qualitative In the Philippines while quantitative in China. In the Philippines, the cytochrome oxidase subunit 1 (COI) genes was used and compared among the species collected and when it was sequenced, it showed differences due to localized gap, mismatch and non-correspondence of bases. And in China, the diversity was analyzed by means of Neiââ¬â¢s gene diversity, Shannonââ¬â¢s information in dex, percentage of polymorphic bands (PPB) an AMOVA anlysis. Both the quantitative and qualitative showed that there was diversification within and among the populations of these snails.INTRODUCTION Genetics is the study of the genes, and the heredity and variation of individuals. Understanding the genome, which is the complete set of chromosomes or the entire genotype of an individual, is important because it helps in the taxonomy of species, especially nowadays wherein advancements in science needs more specific information, and that basing on morphological features is not enough. Life is specified by genomes which contain all the biological information which is encoded in its deoxyribonucleic acid (DNA) and divided into units or the genes.The genes are the blueprint for life because it is the particulate determiner of hereditary traits. Hence, DNA barcoding became a trend for scientists and researchers for the understanding of the different variations in the traits of different o rganisms. The golden apple snails (Pomacea canaliculata) originated from the South America, Central America, the West Indies and the Southern USA (Pain 1972) and was spread in the past decades to the different parts of Southeast Asia, namely Philippines, China, Thailand, Cambodia, Hong Kong, Indonesia and Japan.The introduction of the P. canaliculata without prior studies caused damages to the different plants and it became an invasive species which resulted to becoming pests to humans and competitors to other local snails, example of which are those from the genus Pili. The P. canaliculata was observed to have different growth and reproduction in different parts of Asia, together with their external characteristics due to the different habitats and environmental conditions (Keawjam, 1986 and 1987), therefore there is the possibility to misidentify two sympatric species as one.On the other hand allopatric populations inhabiting different habitats may show ecomorphological variations and questionable species status and it was also suggested that the golden apple snails had high adaptability hence it was easier for them to form new populations (Dong et al. 2011). The understanding of the genomes of different species is a trend for scientists these days but the information about the different mollusks is still limited. The basic information on the number of species and/or population is of help for conservation programs (Carvalho and Hauser, 1994) and for building appropriate management schemes.In contribution, the studies aims are to evaluate the genetic diversity of the golden apple snail population in Asia, namely, Philippines and China via molecularly characterizing the P. canaliculata and to find different ways of analyzing the gathered data from the sequenced DNA of the said species. REVIEW OF RELATED LITERATURE Genetic diversity (Reed 2005) The significance of genetic diversity arose from two necessities: genetic diversity is required for populations to evo lve in response to environmental changes and heterozygosity levels are linked directly to reduce population fitness via inbreeding depression.The amount of genetic variation a population contains is predicted to correlate with current fitness and, in the case of heritabilities (which can remain high or even increase despite severe reductions in population size) with evolutionary potential. This correlation between fitness and levels of genetic variation, however, may be weak or nonexistent due to the neutrality of molecular markers used in estimating heterozygosity, nonadditive genetic variation and the purging of deleterious alleles because of increased selection against homozygotes.There is a body of literature that suggests that allozyme heterozygosity is a good measure of population fitness and adaptive potential. Others caution though that such molecular genetic data generally reflect only a small portion of genome and thus may not be indicator of adaptive genetic differences. But molecular markers may be useful for assessing the extent of genetic drift. Moreover, deleterious alleles, in mutation-selection balance, are responsible for at least half of the genetic variation in fitness.Selection has the tendency to purge the population of the deleterious recessive alleles which in theory creates inbred populations with a higher fitness than their outbreed progenitor. In other words, inbred populations with less genetic diversity would have higher fitness if the population is not kept small enough for a long enough to allow the fixation of deleterious alleles to occur. Fitness and future adaptability are reduced in smaller populations of plants and animals due to drift and inbreeding depression.Commonly used surrogates for fitness such as heritabilities, heterozygosity, and population size, significantly correlate with fitness and explain 15-20 % of the variation in fitness. Correlations suggest that many populations have reduced fitness as a result of inbre eding depression and genetic drift. There is much fuss and concern thus, over genetic variation because of the fact that endangered species typically have lower levels of heterozygosity and the loss of adaptive genetic variation and inbreeding depression puts wildlife populations at an increased risk of extinction.Finally, this increase occurs as a result of the reduction of productive fitness because of inbreeding depression or due to the failure of tracking the change in abiotic and biotic environment of the population as a result of the loss of genetic variation through drift. DNA barcoding (Moritz & Cicero 2004) At the very core, the purpose of DNA barcoding is for large scale screening of one or a few reference genes in order to assign unknown individuals to species and enhance discovery of new species.In the hope of developing a comprehensive database of sequences that will serve as a comparison tool to sequences from sampled individuals, proponents used DNA barcoding. There i s, however, nothing new with DNA barcoding as it is an offshoot of the use molecular markers for the very same purpose except, in DNA barcoding, there is an increased scale and proposed standardization. The selection of one or more reference genes characterizes standardization, with regards to microbial community and in stimulating large scale phylogenetic analyses if of proven value, though whether or not one gene fits all remain to be a question.Presently, most methods of DNA barcoding are tree-based and can fall into two broadly defined classes. One class is the distance-based, wherein it is based on the degree of DNA sequence variation within and between species. This kind of approach converts DNA sequences into genetic distances and then uses these distances to establish identification schemes. It further defines a similarity threshold below which a DNA barcode is assigned to a known or a new species. There is also the mention by several authors of a ââ¬Å"barcoding gapâ⬠, a distance-gap between intra- and interspecific sequences, for species identification.However, the distance-based approach seems to be ill suited as a general means for species identification and the discovery of new species. One reason is that substitution rates of mitochondrion DNA vary between and within species and between different groups of species. The varied substitution rates can result in broad overlaps of intra- and interspecific distances, and hinder the accurate assignment of query sequences. Another class which is the monophyly-based requires the recovery of species as discrete clades (monophyly) on a phylogenetic tree and is used to assign unknown taxa to a known or new species.Similarly, some issues complicate the use of monophyly in a barcoding framework. For example, the long-recognized problem of incomplete lineage sorting will yield gene genealogies that may differ in topology from locus to locus. The recently divergent taxa may not be reciprocally monophyletic due to lack of time needed to coalesce. In addition, the gene trees are not necessarily congruent with species trees, and the monophyly, while a discrete criterion is arbitrary with respect to taxonomic level.Moreover, there is a recently applied new technique that has been proposed as an alternative to tree-based approaches for DNA barcoding, the so called character-based DNA barcode method, which is based on the fundamental concept that members of a given taxonomic group share attributes that are absent from comparable groups. It is the kind of method that characterizes species through a unique combination of diagnostic characters rather than genetic distances. The four standard nucleotides (A,T,C,G) if found in fixed states in one species can be used as diagnostics for identifying that species.This way, species boundaries can be defined by a diagnostic set of characters which can be increased to any level of resolution by applying multiple genes. Presently, character-based DNA b arcode method has been proved useful for species identification and discovery of several taxa. In the view that single-gene sequence should be the primary identifier of species, a contention arises that if that is the case then thereââ¬â¢ll be a real need to connect different life history stages and increase the precision and efficiency of field studies involving diverse and difficult-to-identify taxa.Although the DNA barcoding community has put emphasis on the importance of large-scale sequence database within the existing framework and practice of systematics, it should be bore in mind that DNA barcoding is not the primary answer in resolving the tree of life. Furthermore, as much as the term ââ¬Å"DNA barcodingâ⬠appealing, it implies, however, that each species has a fixed and invariant characteristic. But this kind of implication renders unrest to the minds of evolutionary biologists.In evaluating thus, the promise and pitfall of DNA barcoding, two areas of application should be distinguished: the molecular diagnostics of individuals relative to described taxa and DNA-led discovery of new species. And although there is little doubt that large-scale and standardized sequencing, when integrated with existing taxonomic practice, can contribute significantly to the challenges of identifying individuals and increasing the rate of discovering biological diversity as presented by this study, the general utility of DNA barcoding still requires further scrutiny.PCR (Moore 2005) In rapidly copying a selected template sequence from a DNA mixture in vitro, PCR offers a wide range of applications such as sequence detection and isolation for research, forensics and species identification through the PCR itself and in combination with other techniques. PCR's new technique uses flourescent probes to monitor the amounted product at end of every cycle and PCR machines look for the cycle at which the can readily detect flourescence.PCR is also being used to monitor RNA through the addition of reverse transcriptase enzyme at the beginning to generate DNA template. In addition, there are now new applications of PCR like single nucleotide polymorphism detection and screening. Cytochrome Oxydase subunit 1 (COI) (Buhay 2009) COI plays a significant role in documenting biodiversity and remains to be the choice for phylogenetic and phylogeographic studies. COI is a mitochondrial protein-coding gene which is a widely accepted marker for molecular identification across diverse taxa.Mitochondrial DNA (mtDNA) have a relatively fast mutation rate, thus they result in significant differences between species. With respect to this, the mitochondrial cytochrome c oxidase subunit I (COI) gene with ~700bp was proposed to be a potential barcode or marker for molecular identification across various taxa. Furthermore, COI is a protein coding gene that has an open reading frame and in thecase of barcoding, COI can be highly divergent from the actual COI sequences which may cause major problems because species identification is based on sequence similarity.Pomacea canaliculata (Cowie 2002) The Pomacea canaliculata belongs to the family Ampullariidae. Its structure appears to have a slight dimorphism in shape of aperture and operculum. Females have broader mouth and concave operculum while convex in male. In terms of reproduction, oviposition often takes place at night or at early morning or evening about 24 hrs after copulation up to two weeks after mating (occurs three times per week) which occurs anytime of the day or night although there may be some diurnal rhythm.On each oviposition occasion a single clutch is laid of highly variable egg number. Moreover, the interval among successive ovipositions for p. canaliculata has been reported to be about five days and hatching generally takes place about two weeks after oviposition. The P. canaliculata breeds only during summer and grows into maturity in less than two months. P. canaliculata is s aid to be prolific and hence has rapid succession of generations which leads to rapid population expansion.They relatively inhabit still water and in water temperatures above 32 degree Celsius, it has been observed that the mortality of p. canaliculata is high. Whereas in low temperature p. canaliculata can survive 15-20 days at 0 degree Celsius, 2 days at -3 degree Celsius but only 6 hrs at -6 degree Celsius. And it is sufficiently tolerant of sea water to survive long enough to be carried by currents from one stream mouth to another, thereby expanding its distribution. P. canaliculata shows preferences among food plants.Its rate of growth has a direct correlation with its feeding on the preferred plant. Moreover, it is able to detect its food plants from some distance using chemical cues in the water. P. canaliculata, however, appears to be relatively generalist and indiscriminate that it is viewed to be particularly voracious compared to other Ampullariids. METHODOLOGY Sampling S nail samples were identified and collected from 2 countries in Asia, specifically in the Philippines and in China, where the P. canaliculata was introduced. In the Philippines; Los Banos (Dong et al. 011, p. 1778), 2 barangays in Tarlac (Brgy. Cabayaoasan, Paniqui and Brgy. Pance, Ramos) and Iloilo (Chichoco & Patdu 2012, p13), 44 snail samples were collected. And in China, specifically from Yuyao and Taizhou in Zhejiang province, Fuzhou in Fujian province, Guangzhou in Guangdong province, Nanning in Guangxi province, Kunming in Yunnan province, wherein a total of 120 samples were identified with the conserved sequence by Matsukura et al. (2008) and Pan et al. (2009) and then was collected (Dong et al. 2011, p. 1778).The snails were then stored, either by wrapping in paper, freezing or preserving it in ethanol, and brought into the respective labs in each country for the next steps; DNA extraction, PCR, electrophoresis and sequencing. DNA extraction The two studies used the phenol-c hloroform method (Bergallo et al. 2006) with an alternative of the Qiagenââ¬â¢s Dneasy extraction kit for China. The DNA concentration was determined spectrophotometrically and adjusted by a mini-gel method (Maniatis et al. , 1982) when the extracted DNA was enough, it was stored at 4oC to -20oC until needed. PCR and ElectrophoresisThe PCR method was basically done by choosing the right primers that will yield clearly reproduced bands and they tested the proper amounts and amplification effects of the components of PCR, which were the Mg2+, dNTPââ¬â¢s, DNA templates and polymerase, and the primers. After the mixture of the components and the DNA extracted, it was carried out in the thermocycler programmed for pre-denaturing at 94à °C for 3 min, followed by 26 cycles of 94à °C for 10-30s, 36-52à °C for 30-45s, extension of 65-72à °C for 60-90s, and the final extension for 5-7mins at 72à °C for final extension with 38-48 cycles.After which, the amplified products together w ith negative controls were run in electrophoresis to be separated and tested for contaminations, respectively. The products were then purified later on with the respective kits present in each lab. In the Philippines, the reaction was done with 2? L MgCl2, 5? L PCR buffer, 1? L dNTP, 2. 5 ? L of the primers, which were the LCO1490 and HCO2198, distilled H2O with 22. 75 ? L, 0. 25 Taq, and 10 ? L Q-buffer. The electrophoresis was done after the ethidium bromide staining (Maniatis et al. , 1982), analyzed through 1. % agarose gels and visualized under a transilluminator. In China, they made use of the ISSR-PCR analysis where they got four primers, which produced clearly reproduced bands, out of the 90 that was screened from the University of British Columbiaââ¬â¢s primer set and the reactions were done with a volume of 20 ? l, containing 0. 2 mM of each dNTP, 1. 5 mM MgCl2, 0. 5 ? M primers, 1 U Taq polymerase and 10 ng DNA template, and also with the determination of the optimal r eaction system of ISSR for P. canaliculata (Dong et al. 2011, p. 1779).The productsââ¬â¢ sizes after the amplification was estimated using DNA marker DL2000 and then was run in electrophoresis, which was done on 6% polyacrylamide gels, visualized by silver staining and then photographed (Li et al. , 2009). Sequencing/ Data analysis Chichioco and Patdu (2012) sent the DNA samples to the First Base Laboratory in Singapore for sequencing and the results were sent back to the DNA barcoding Laboratory after a week. The COI sequences were aligned in the BLAST, specifically the STADEN package version 1. 5. 3 and Bioedit Sequence Alignment Editor version 7. 0. 9. 0.Aside from the sequences sampled, other sequences and their haplotypes from the GenBank were also compared and matched. In Dongââ¬â¢s (2011) research, he made use of the RAPD fragments by labeling them into binary matrices, used them to get the similarity index, Sxy = 2nxy / nx+ ny, where nx and ny represent the number of R APD bands in individuals x and y, and nxy represents the number of shared bands between individuals, as stated by Nei and Li (1979), then averaging it across all the possible comparisons between individuals within a geographic sample to get the within samples similarity (Si).Between sample similarity corrected by within sample similarity Si and Sj of geographic samples i and j, respectively) is also calculated between pairs of individuals across samples i and j using the equation; Sââ¬â¢ij = 1 + Sij ââ¬â (Si and Sj)/2. Genetic distance between paired samples was then calculated as Dââ¬â¢ij = 1- Sââ¬â¢ij (Lynch, 1990). RESULTS AND DISCUSSION In the Philippines (Chichioco & Patdu 2012, p. 18-31) The collected samples from Brgy. Cabayaoasan were found in the elevated parts of a rice paddy, specifically, it was a muddy substrate with decaying leaves from the rice plants and surrounding trees while those that was found in Brgy.Pance was in the muddy bottom of the shallow fis h pond in the roots of water lilies and grasses. The samples from the two barangays in Tarlac and Iloilo had relatively different colors and sizes. Those that were collected from Brgy. Cabayaoasan had the largest size and they are colored black while those in Brgy. Pance had brown in color and still, those in Iloilo had very small sized specimens and some of the specimens can be mistaken as Pila conica snails if not examined properly. Primers affect the amplification success greatly, since according to Hajibabaei (2005) a 95% success is necessary for barcoding.The primers LCO1490 (SENSE) and HCO2198 are generally used for the amplification of forward and reverse fragments from COI genes. The DNA samples were subjected to the PCR and agarose gel electrophoresis (AGE), and they produced single discrete bands that suggest that the fragments were homogenous and start and end at the same point (Reece 2004). The bands that were brighter and distinct are more appropriate for sequencing bec ause it means that the DNA fragments were well amplified. The best DNAââ¬â¢s were chosen and forwarded to the First Base Laboratory in Singapore for sequencing.At the return of the results, other sequenced DNA barcodes were also collected and was aligned and compared with the Basic Local Alignment Search Tool (BLAST) database. Pance, IICK & IIPC1,3 from Iloilo) from the Philippines using Bioedit Sequence Alignment and ClustalW multiple Alignment (Chichioco & Patdu 2012, p. 26) By aligning the sequenced data, it can be seen that there are both similarities and differences among the genetic make-up of the samples.The differences are due to localized gap, mismatch and non-correspondence of bases along the COI fragments as pointed out in fig. 2 Fig 2. Comparisons of the COI sequences of the P. canaliculata samples from 35bp- 120bp (Chichioco & Patdu 2012, p. 28 ) As emphasized in fig. 2, on the 55bp-58bp, a sequence from CPT1 was observed having (5ââ¬â¢-AATT-3ââ¬â¢) while all the others have (5ââ¬â¢-GGTA-3ââ¬â¢). Even though this is a noticeable difference and could have been caused by mutation or variation, the difference is still low enough and less that 1% difference to be considered significant.But on the other hand, the PCa1 sample had 36 different base pairs compared with the others, which was 5. 5% difference and is high enough and can be considered significant since it is ;4% divergent(Meyer and Paulay 2005). Then with a ~98% confidence, it could be said that PCa1 is from an independent evolutionary lineage and might indicate a divergence within or outside its population or might have occurred due to relationships and interactions among the other species.The introduction of the P. canaliculata to different places may have an effect on its intra- and interpopulation and might be why it has various genetic sequences altho ugh it goes against the theory that introduced species becomes a founding population in a new habitat thus they have a limited gene pool and as a consequence genetic drift, which removes variability since it affects all genes, and bottleneck might occur, which reduces the new species to have a reduced genetic diversity.To observe the genetic variability, the sequences collected were compared with those from GenBank with the use of the BLAST software. As a result from 81 COI barcodes and 55 haplotypes, the samples collected showed 99% and 100% similarities with the different haplotypes thus it showed that the species has a high diversity within the populations. The phylogeography within and among the species does not apply on the P. analiculata since intra- and interpopulation diversity was observed which was shown by the multiple introduction throughout the Philippines, hence the different times of the introduction contributed more to its diversity and it coincides with the migrant pool model that says that the introduced population acquires more genetic variability because of the multiple sources of genetically divergent populations as compared to that of the local species (Slatkin 1997, Sakal et al 2001). In China (Dong et al 2010) The chosen primers an average of 124. bands, since they generated a total of 498 bands, which ranged from 150-2500bp and qualifies them for barcoding, as seen in table 1. Among the 140 individuals, 435 bands were polymorphic which was different for each primer. In table 2, Neiââ¬â¢s gene diversity (H) varied between 0. 2612 and 0. 3340, with an average of 0. 3044, and arranged in a descending order the populations, LB ; KM ; NN ; FZ ; TZ ; GZ ; YY while the Shannonââ¬â¢s information index (I) ranged from 0. 3910 to 0. 4856, with an average of 0. 4499.At the species level, the values of Neiââ¬â¢s and Shannonââ¬â¢s showed the same trend as that of PPB. AMOVA analysis showed that there are highly significant (P ; 0. 001) genetic differences among the seven populations of P. canaliculata. The genetic diversity was mostly due to the differences within the population (92. 76%) while the rest was due to among populations. The analysis tells the same as that of the Neiââ¬â¢s and Shannonââ¬â¢s information, which says that there was a relatively high level of genetic differentiation among populations. CONCLUSIONGenetics of different species are studied by means of DNA barcoding, mostly of the COI gene in the mitochondria, to know the taxon of a species and to understand their trends and characteristics not only morphologically but also genetically. The diversity of a species can also be tested by means of DNA barcoding as seen in the study of the Pomacea canaliculata. The P. canaliculata was introduced in Asia for agricultural purposes and was seen for its benefits but not its drawbacks, which later on resulted to it being invasive and a pest for both humans and other species.To understand the P. can aliculata further, its diversity was studied by means of DNA barcoding and was analyzed both qualitatively and quantitatively in the Philippines and China, respectively. Both the analysis showed the same outcome, wherein the results showed high levels of genetic diversity among populations. Because invasive species tends to give a negative feedback to those species in the local area, it is important to understand these alien species and to know how diverse they are so that proper management of these species could be done. Advancing Technology
Maintaining the Role of Technology Technology has been and will forever be advancing. It plays an important role in everyone's life, and can be found anywhere. Whether it's a six year old child playing with an IPhone, or an 80 year old lady reading a book on her kindle. There is Just no hiding from it. But with the advancement of technology, is society advancing with it, or Just becoming robots? To succeed in not being taken over by technology people must be true to themselves, set limits, and detach from it. Now a days, almost everyone has some sort of social media network.It could be a Twitter, Facebook, Instagram, etc. That is what makes the internet such a dangerous place. While on the computer anyone could put anything as themself. They could make fake profiles or give false information about themselves, and the only person who would know is them. Someone could be talking to who they believe is a cute 20 year old girl, who they want to meet up with and date, but in reality it co uld be a 30 year old trucker playing a Joke. It is easy for people to hide behind a computer screen and that is why they do it so often. A prime example of this can be found from the T. V show ââ¬Å"Catfish. What happens in the show is, two people end up talking to each other nline; they start talking for a while, and then they fall in love without ever seeing each other. Then the hosts of the show go to one of the people and help them meet the ââ¬Å"love of their life. â⬠But out of the 24 aired episodes, only two people have actually been who they really said they were. But there are many more people who aren't being who they really say they are. So while using technology people should be who they really are, and Just because they can hide behind a computer screen, they should still be true to themselves. Hiding from technology is not an easy task.Some people actually need technology or important things, so they can't Just throw it away. So instead of shutting out technolog y completely, people should Just set limits for themselves. The limit is up to them; either hardly using technology and Just going outside and enjoying the great outdoors, or cutting back to a certain amount of hours they use it. When setting these limits, take into account the time that you really need it. For instance, for work, checking important e-mails, or even school. People should cut back and use it when they actually need to, instead of using it all of the time when they have nothing better to do.Using technology for entertainment is alright, Just try to cut back and set limits so that it isn't over used. Just because someone doesn't check their Facebook every minute, or stare at their phone while their Twitter news feed goes crazy doesn't leave you out of the loop. And that is the key to not being taken over. A big problem that people get is that they feel that they must constantly check their things so they don't miss something important. The problem is that they are miss ing out on what is happening right now and not focusing on the current. They could be driving and get an urge that they'll miss a party invite, so they must check.Or if they are at a family get together and they are bored, they will Just go on Facebook and think that, that is a better way to spend their time, but they actually shouldn't be using their phone, and instead making the best of should detach from technology and the virtual world. No matter the age, no matter the gender, everyone uses technology. It helps people get through their everyday lives. It could be used for business or Just entertainment, but no matter what the reason is people should always look out to not be used by technology. To do so they should remember to be true to themselves, set limits, and to detach from technology.
